Pretreatment of Shaoyao-Gancao-Tang alters Pharmacokinetics of Intravenous Pacli

Objective: To investigate the effect of Shaoyao-Gancao-Tang (SGT) on the pharmacokinetics of intravenously administered paclitaxel in rats. Method: Paclitaxel were administered to rats intravenously (3 mg/kg) with or without the concomitant administration of SGT(752 mg/kg, single day or 14 consecutive days pretreatment). And quantification of paclitaxel in serum was achieved using a simple and rapid UPLC method for pharmacokinetic study. Result: Pharmacokinetic parameters of paclitaxel were significantly altered with 14 consecutive days pretreatment of SGT. There were significant decreases in area under the curve (P<0.01) and the total clearance increased by 1.2 fold (P<0.01)in 14 consecutive days pretreatment of SGT. The pharmacokinetic parameters of paclitaxel were not significantly affected by single day pretreatment with SGT. Conclusion: Administration of SGT for 14 consecutive days increased the metabolism of paclitaxel in this study, and 1-day pretreatment had little effect on pharmacokinetics of paclitaxel. The results will add important information to the study of interaction between TCMs and chemotherapy and also help in better use of SGT in adjunctive therapy of cancer patient.
