Gypsophila saponins inhibit proliferation and promote apoptosis of gastric cance

Objective:The purpose of the study was to evaluate the anti-proliferative activity and apoptotic effect of Gypsophila saponins on human gastric adenocarcinoma SGC-7901 cells and its mechanism. Method: MTT assay to determine cell viability and to calculate the IC50; Apoptosis Dection and Cell Cycle Analysisy by Flow Cytometry; To detect the expression of Wnt2 and Wnt2B mRNA by Quantitative Real-time PCR. Result:MTT assay showed that Gypsophila saponins inhibited the proliferation of SGC-7901 cells in a time- and dose-dependent manner. Apoptosis dection and Cell cycle analysis respectively indicated that apoptotic SGC-7901 cells significantly increased in a dose-dependent manner after treatment with Gypsophila saponins and Gypsophila saponins treatment in 24h arrested most cells at G0/G1 phases and significantly reduced cells at S phase in a dose-dependent manner (P<0.05). Quantitative RT-PCR showed that with the increased concentration of Gypsophila saponins, the expression of Wnt2 and Wnt2B mRNA was down-regulated in a dose-dependent manner. Conclusion: In conclusion, Gypsophila saponins inhibit gastric cancer cell proliferation and promote its apoptosis through down-regulation of Wnt genes expression. Thus, Gypsophila saponins serve as a potential therapeutic agent for the treatment of gastric cancer.
